The effect of 5-azaC on global DNA methylation levels in Escherichia coli. E. coli wild-type strain BW25113 was grown in LB at 37 °C in the absence of 5-azaC, the presence of 0.5 μg/mL 5-azaC, 5 μg/mL 5-azaC, and 50 μg/mL 5-azaC. At two hours (logarithmic phase, L) and eight hours (early stationary phase, S), genomic DNA was isolated. DNA was incubated in the absence of restriction enzyme, presence of BstNI (B) (not blocked by Dcm methylation), or presence of PspGI (P) (blocked by Dcm methylation). DNA samples were analyzed by agarose gel electrophoresis and ethidium bromide staining
